We proposed a new strategy to control the drug release rate of polymersomes by controlling membrane phase separation. This was achieved by employing membrane-forming blocks with different molecular architectures and cross-linking degrees. This work shed light on the design of programmed drug release vehicles.
In this study an easy-operating but effective strategy is used to prepare PVal which can self-assemble into nanoparticles (PVal-NPs, ~104 nm) by nanoprecipitation and have a higher drug payload for tumor chemotherapy. This nanoplatform is stable under physiological conditions and could induce the apoptosis of 4T1 cells like free DOX while alleviating its toxicity. The in vivo evaluation indicated that DOX@PVal-NPs have a high tumor accumulation, penetration and therapeutic performance after intravenous injection without causing toxicity to main organs. Overall, this study provides a new strategy for synthesis of poly(amino acid)s as drug carrier to deliver antitumor drugs.
By using a dinuclear salen-Cr(III) complex, copolymerization of PO/CO2 and LA could be realized more successfully than that of mononuclear salen-Cr(III) complex. So that, gradient and random copolymers were obtained with modificable microstructure by adjusting the reaction temperature.
we here report a new methodology for tuning the saline stabilities of polyplex nanovesicles of oligonucleotides by modifying the polycation with Zinc (II)-coordinative cationic chains.
We design a fused in sarcoma (FUS)-derived protein by fusing a hexhistidine-tag to the low complexity domain of FUS, which undergoes liquid-liquid phase separation in the presence of metal ions. Regulation of dynamic coacervation of FUS with the competitive binding strategy offers a possibility to design protocells as a controlled release system.
We developed a "gradient deposition" technique to coat a continuous and smooth layer of chitosan onto the surface of absorbable sutures. The coated sutures exhibited excellent anti-bacterial and anti-inflammation activities for significantly enhanced wound healing effect.
Inspired by natural heat shock protein, we fabricated a self-assembly nanochaperone based on mixed-shell polymeric micelle (MSPM) as a novel tau-targeted AD therapy. With tunable phase-separated microdomains on surface, the nanochaperone could effectively bind with PHF6 aggregates, inhibit PHF6 aggregation, block neuronal internalization, thus significantly alleviating PHF6 mediated neurotoxicity.
We propose a cationic block polymer based on polyethylene glycol and poly aspartic acid for siRNA delivery. The vector exerts multi properties including good biocompatibility, high encapsulation efficiency, and tailorable modification for superparamagnetic iron oxide and targeting components, which result in a highly efficient and tumor targeted siRNA delivery.
MSCs exhibit rapid proliferation and differentiation potential maintenance on microcarriers with intermediate rigidity which stimulates the fastest spreading rate.
Herein, we develop a new strategy in which photoacoustic (PA) imaging reagents with better properties can be easily optimized by polymerization. The polymerization strategy can effectively promote the PA signal by specifically increasing the thermal-to-acoustic conversion efficiency. The polymers with intense intramolecular motion exhibit an amplified PA signal by elevating thermal-to-acoustic conversion and its higher light-harvesting capability at redshifted region. This work highlights a simple and available design guideline of polymerization for amplifying the PA effect and optimizing existing materials.
We developed an ROS-responsive PEGylated polypeptide-based macromolecular ethyl caffeate (EC) prodrug via phenylboronic esters for the alleviation of uveitis. The antioxidative 4-hydroxy benzyl alcohol (HBA) and EC can be released under the stimulation of ROS, which can effectively protect cells against oxidative stress-induced injury in an ROS-depletion way.
Inspired by bio-functions and microstructures of extracellular matrixes and mussel foot proteins, we constructed the glucose-/catechol-functionalized ε-polylysine-based hydrogels, which presented excellent hemostasis and wound healing performance, and the hydroproline level within 14 days is comparable to that in normal skins. The cooperative glycosylation and catechol-modification make commercial ε-polylysine suitable for constructing high performance wound hemostats and dressings with potential transition applications.
In this work, T-cRGD NPs was obtained by the self-assembly of DSPE-PEG-cRGD and TPP-PPh3-4PEG for targeting sonodynamic therapy. Peptide targeting endows T-cRGD NPs with superior ultrasound therapeutic efficacy and tumor enrichment ability.
