Chun Yang, Tong Wu, Min Sun, et al. Thioether-based Semi-permeable Polymer Vesicle for
Hyperuricemia is a major risk factor for gout
renal injury
and cardiovascular disease
necessitating effective control of serum uric acid levels. Existing therapies face challenges such as limited efficacy
renal toxicity
and dosing complexities. Urate oxidase (UOx) offers a potent enzymatic degradation strategy but suffers from
in vivo
persistence
immunogenicity
and the generation of the harmful byproduct hydrogen peroxide (H
2
O
2
)
which limits its clinical utility. Here we report a dual-functional polymeric vesicle that functions as a microreactor for UOx delivery. The vesicular membrane was designed to reduce the direct exposure of encapsulated UOx to immune components
while thioether groups embedded within the membrane rapidly scavenge H
2
O
2
generated
in situ
. This integrated design enables simultaneous uric acid degradation and byproduct elimination without introducing additional enzymes. UOx@vesicle retained 78.1% of the enzymatic activity of free UOx. In hyperuricemic mice
UOx@vesicle maintained a significant uric acid-lowering effect for up to 73 h
approximately 2 days longer than free UOx
while attenuating immune responses and decreasing serum biochemical markers associated with hepatic and renal dysfunction. This work provides a general strategy for enhancing the safety and efficacy of enzyme-based therapeutics for hyperuricemia and other reactive oxygen species-related diseases.
